• supplemental material - Movie 2 Peripheral growth cone retracts from Sema3D. Two branches of a peripheral axon are labeled. The branch on the left contacts an ectopic Sema3D-GFP expressing cell (green) and retracts from it while the other branch of the same cell is advancing steadily forward. The diffuse green is out of focus fluorescence. Images were captured every two minutes, total time shown is 124 minutes. Lateral view, anterior left.

A peripheral axon retracts after contact with ectopic Sema3D-GFP and at the same time back branches form along the axon shaft. • supplemental material - Movie 5 TAGMO dramatically affects central growth cone behavior. The first clip shows two wild type central growth cones extending along each other. They make steady forward advance over a total time of 157 minutes. The second clip shows a central growth cone in a TAGMO injected embryo. The growth cone has not extended far from the cell body and makes no significant forward advance during 212 minutes total time. Note that the growth cone remains highly active throughout this period.
