Supplemental Movie: Three-Dimensional Reconstruction of Degenerated Neuron with Intraneuronal Aβ Seeding an Amyloid Plaque.
A three-dimensional reconstruction of a confocal Z-stack of images of the Tg6799 brain section shown in Fig. 5G was generated
using the software program Velocity. The reconstructed image was then rotated along three spatial axes and saved as an AVI
movie file. The reconstruction shows an intraneuronal Aβ-containing neuron (center) with an amyloid plaque (anti-Aβ R1282
antibody, red) emerging from the upper left part of the neuron. This neuron displays a shrunken, disrupted morphology and
has very little anti-NeuN antibody staining (green), indicating severe neurodegeneration. In addition, other neurons in the
section appear to be in various stages of degeneration (e.g., lower right) and may also be seeding amyloid plaques (e.g.,
far left center). Note the presence of Aβ-filled neurites passing through the section (red linear structures running at a
diagonal) and the amyloid deposit at far right center. The Tg6799 brain section was prepared and co-stained with anti-Aβ (R1282,
red) and anti-NeuN (green) antibodies, as described in the legend to Fig. 5 and Materials and Methods.
