ABC treatment in the mPFC does not block cocaine cue-induced reinstatement in the absence of memory reactivation, nor does it block sucrose cue-induced reinstatement after a novel VR5 memory reactivation session. A, Active lever presses during the last 3 d of training in rats trained on an FR1 schedule (no-react and sucrose groups) or an FR3 schedule (FR3; Fig. 2). As expected, there was an increase in lever pressing in the FR3 versus no-react group trained on an FR1 schedule. B, The number of cocaine infusions was not different across the last 3 d of training. C, The number of active lever presses for sucrose pellets was not different across the last 3 d of training (trained on an FR1, reactivated on a VR5). D, The number of sucrose rewards was not different across the last 3 d of training. E–H, During memory reactivation, rats trained on the FR3 schedule were also given an FR3 schedule for memory reactivation, while rats trained for sucrose pellets were given a VR5 schedule for memory reactivation. In the FR3 groups, active lever presses (E) or cocaine infusions (F) were not different between Veh and ABC groups, and in the sucrose VR5 groups, neither active lever presses (G) nor number of sucrose pellets (H) were different between Veh and ABC groups. I–K, Active lever presses during extinction in no react (I), FR3 (J), and sucrose VR5 (K) groups. There was a decrease in responding across the session for all three groups, with no differences between Veh and ABC groups. L–O, During cue reinstatement, there were no differences between Veh and ABC groups in active lever presses (L) or cue rewards (M) in the no-react and FR3 groups. There were also no differences between Veh and ABC groups in active lever presses for sucrose (N) or sucrose pellets (O) in the sucrose VR5 group. P, Q, Active lever presses during PR schedule over 3 consecutive days in no-react, FR3 (P), or sucrose VR5 (Q) groups. There was only a decrease over day for active lever presses and sucrose rewards in the sucrose VR5 group. R, S, Microinjection sites for no-react, FR3 (R), or sucrose VR5 (S). Gray dots, Veh; red dots, ABC. Data are mean ± SEM. No react, Veh N = 7; ABC N = 6; FR3, Veh N = 5; ABC N = 6; sucrose VR5, Veh N = 9; ABC N = 5. Extended Data Figure 3-1 shows the number of inactive lever presses and rewards from animals shown in Figure 2. Extended Data Figure 3-2 shows statistical comparisons for Figure 3.
