Figure 8.

The inhibition of LINC complex activity mimicked most of the phenotypes induced by MTCL2 knockdown. A, Schematic of our working hypothesis. During the late phase of CGN polarization, the MTCL2-mediated association between the Golgi apparatus and the MT cage induces an upward extension of the Golgi apparatus along the nucleus. This recruitment to the MT cage results in the sequestration of the Golgi apparatus from the roots of the preexisting bipolar dendrites. The molecular organization of the LINC complex on the nuclear membrane is illustrated on the right. B, A typical cross-sectional view of CGNs expressing GFP-KASH1, fixed at DIV6, and stained for MTCL2 and GS28. The white dotted lines indicate the focus plane from which the confocal image shown in Figure S8A was obtained. Scale bar, 5 µm. C, Apical confocal section of a GFP-KASH1-expressing cell stained for MTCL2 and tyrosinated tubulin. Scale bar, 5 µm. D, θ distribution of the Golgi position in CGNs expressing GFP or GFP-KASH1, examined at DIV6. Data are presented as polar histograms as shown in Figure 3E for the indicated number (n) of cells from three independent experiments. E, Height distribution of the Golgi apical edge in the same CGNs analyzed in D. F, θ distribution of individual dendrites of CGNs expressing GFP or GFP-KASH1, examined at DIV6. Data are presented as polar histograms as shown in Figure 3B for >400 dendrites of the indicated number (n) of cells from three independent experiments. G, H, Distribution of the number of dendritic tips (G) and averaged dendritic length (H) of the same CGNs analyzed in F. Each data point in E, G, and H represents the result of a single neuron. The means ± SE are shown in D and F, whereas the means ± SD are shown for each scatterplot (E, G, H). The Wilcoxon rank-sum test was used to examine statistical significance.
